Her breakout role as Brenda Walsh in the Aaron Spelling-produced TV series Beverly Hills, 90210 in 1990 brought her worldwide exposure and acclaim. Brenda got her a Young Artist Award nomination for Best Young Actress Starring in a Television Series in 1991 and 1992. In 1994, Doherty exited the show after the fourth season.
After that, her career was mostly made-for-TV movies, though she did have a lead role in Kevin Smith’s 1995 film Mallrats and later appeared in Jay and Silent Bob Strike Back. Spelling cast her in another of his television shows, Charmed, in which she played Prue Halliwell, the oldest of three witch sisters. During the second and third seasons of the show, Doherty also directed a few episodes.
She joined the cast of the Beverly Hills, 90210 spin-off for The CW Broadcast Network in 2008, 14 years after her final television appearance as Brenda Walsh, for a rumoured $40,000–50,000 per episode. She appeared in four of the first six episodes of the new series as a guest star, recreating her former role of Brenda.
Her character, who is now a successful theatrical performer and stage director, returned to direct the high school musical as a guest director. Doherty mentioned that she was open to returning to the series later in the season after her initial guest appearance.
Doherty was confirmed as a celebrity contender on Dancing with the Stars for the tenth season on March 1, 2010. On Monday, March 22, 2010, the season premiered. In his sixth season on the show, Doherty was paired with two-time champion Mark Ballas, however the two were the first couple eliminated in the second week on March 30.
“If we had just done scores,” the judges added, “Doherty wouldn’t have departed.” She wanted to go on Dancing with the Stars to show her ill father how proud she was of him. For the finale, Doherty reappeared.

Following Perry’s death in March 2019, the Riverdale team reached out to his longtime friend Doherty, who agreed to appear in the season 4 Perry tribute episode because Perry had been pushing for her to make a cameo since the first season.

Doherty was diagnosed with breast cancer for the first time in 2015. Her breast cancer had gone into remission by 2017. She was 46 at the time and had to seriously consider whether or not she wanted to start a family. Chemotherapy, radiation, and surgery for cancer took a toll on her health, according to Health, but her age also had a role.
Then, in 2020, Doherty was struck yet another setback. Her cancer had returned, this time in Stage 4 and metastatic, meaning it had spread beyond her breasts and lymph nodes. She told Elle at the time that she felt like she had a lot more time.
